Glucose reagent: Combine 30 ml of O-Toluidine and 470 ml of glacial acetic acid. Mix well.
Standard solution: Accurately weigh 100.0 mg of Dextrose Monohydrate WS into a 100 ml volumetric flask, bring to volume with purified water and mix thoroughly. Pipette 10.0 ml of the solution in 50 ml volumetric flask and bring to volume with purified water to obtain a final concentration of about 0.2 mg dextrose monohydrate per ml.
Sample
solution: Transfer
sample equivalent to 100 mg of Dextrose Monohydrate in 50 ml volumetric flask
and bring to volume with purified water. Pipette 10.0 ml of the solution in 100
ml volumetric flask and bring to volume with purified water to obtain a final
concentration of about 0.2 mg per ml.
Procedure: Transfer 3.0 ml of each standard solution, sample solution and blank into test tube. Add 15.0 ml of glucose reagent to each test tube. Place the test tube in a beaker of boiling water for 10 minutes, (Note: Solution is stable for 60 minutes after boiling). Cool the test tube to room temperature. Zero the spectrophotometer at 630 nm using the blank.
Take the absorbance of the standard
solution and sample solution at 630 nm.
